PostDoctoral Fellow - Developing Immune Cell Screening

Sanger Institute

Do you want to help us improve human health and understand life on Earth? Make your mark by shaping the future to enable or deliver life-changing science to solve some of humanity’s greatest challenges.

About the Role:

We have an exciting opportunity for a Postdoctoral Fellow to join Vento-Tormo Lab in our Cellular Genetics Programme for a 3 year contract.

You will be working on an extensive collaborative project (Open Targets) with other academic and industrial partners to develop immune cell screening platforms in order to target a large number of genes. We will use established CRISPR/Cas9 technology, but also develop new strategies to screen and multimodally assess gene perturbations in in vitro-derived immune cells. With this comprehensive approach, we will gain insight into the effects that gene perturbations have on transcriptional effects and their role in gene regulatory networks in the setting of different immune cell populations. This will inform target prioritisation in drug development processes.

This project will involve tight collaboration with members of other teams working on this collaborative project, both experimental and computational.

You will be responsible for:

Your position will be in Vento lab, where you will work closely with a Research assistant on setting up iPSC-derived macrophage genetic screens. You will work closely with other groups on the project, including a Gene editing group. You will take the scientific lead on optimising the existing, but also establishing novel gene editing approaches in macrophages on a high throughput level. This project will involve close collaboration with a computational postdoc to analyse and interpret the data and to present the data to both academic and industrial partners.

About You:

You have experience working with iPS cells, large-scale genomic data generation and CRISPR screening. You are independent, but comfortable working as a part of a bigger team. We welcome your application if you have skills and experience in the following:

  • Stem cell culture, especially on a high throughput scale
  • Gene editing (CRISPR/Cas9) and screening
  • Cellular assays
  • Single cell RNA sequencing
  • Project management skills
  • Immunology

Essential Skills:

  • PhD in molecular biology, biotechnology, genetics, immunology or a similar discipline with a publication record
  • Proven knowledge of pluripotent stem cells (iPSC and ESC)
  • Proven knowledge of gene editing technology (CRISPR/Cas9) and screening processes
  • Experience with CRISPRi and CRISPRa technology
  • Practical knowledge of single cell-omics (e.g. scRA-seq)
  • Experience in carrying out both independent and collaborative research
  • Ability to plan and prioritise own work in order to meet deadlines, including using initiative to plan research programmes
  • Highly developed communication skills to engage effectively with a wide-ranging audience, both orally and in writing, using a range of media
  • Commitment to personal development and updating of knowledge and skills
  • Working collaboratively with others and building working relationships with stakeholders at all levels
  • Demonstrates inclusivity and respect for all

Desirable Skills:

  • Knowledge of immunology, especially macrophage biology
  • Experience with iPSC differentiation protocols
  • Experience with cellular assays (phagocytosis)
  • Knowledge of target identification processes
